Summary:
Atherosclerosis, the underlying cause of coronary artery disease that is the
leading cause of death in most parts of the word, is a multifactorial disease.
Several mechanisms participate in its initial manifestation and progression,
mainly involving the triad: hyperlipidemia, oxidative stress, and inflammation.
Specifically, evidence suggests that oxidative modification of LDL,
hypercholesterolemia/hyperlipidemia and the chronic inflammatory responses,
between bloodstream components and the arterial wall, could synergistically
lead to the atherosclerotic plaque formation.
Since atherosclerosis is a multifactorial disease, different
pharmacotherapeutic approaches have been applied. According to this strategy
and based on the pathogenic mechanisms involved in the onset and progression
atherosclerosis, we designed compounds that combine antioxidant, SQS inhibitory
as well as anti-inflammatory properties. Thus, we synthesized and evaluated
five series of compounds and carried out structure-activity relationships.
