Summary:
Osteoarthritis (OA) is the most common chronic disease of the musculoskeletal
system, affecting the majority of the elderly and several young people.
According to the World Health Organization (WHO) OA is one of the most common
causes of disability and gender and socio-economic costs of OA is huge.
OA is a multifactorial disease. The genetic component of OA brings great
scientific interest as genetic influence on OA is in the range 35-65%. Some of
the correlations for OA is higher among siblings than between parents and
offspring. Studies of twins, siblings and families with both parents suffering
of OA showed that the familial occurrence of the disease is strongly associated
with genetic factors (heredity likelihood for a osteoarthritic knee is 40%,
while the articular ends of hand and hip are 65%). The genes implicated in OA
is not specifically a hinge, thereby enhancing the theory of multivariate -
multigene nature of the disease. This fact and the late onset of the disease
make it binding studies and identification of candidate genes very difficult.
The evolution of the science of genetics and the mapping of the human genome,
have allowed for more complex genetic studies.
In recent years, there has been great interest in the search for genes that
predispose to disease studies mainly with linkage analysis and association
studies of particular genes.
