Μελέτη της έκφρασης των υποδοχέων της ορμόνης CRH, CRH-R1 και CRH-R2, σε έμβρυα ποντικού 2-, 4- και 8-κυττάρων
Summary:
The purpose of this study is to quantify the expression of both CRH
receptors, CRH-R1 and CRH-R2, in mouse embryos that are in the stages of 2-, 4-
and 8-cells.
For the realization of this study were used female mice 5-6 weeks old.
Originally induced ovulation and then followed in vivo fertilization by male
mice 3 months old. Then we sacrificed the mice, removed the fallopian tubes,
removed the embryos and cultured them in a suitable culture media until they
reached at the stages that we wanted. Followed isolation of the total RNA
(RNA-Extraction) for the embryos of 2-, 4- and 8-cells, conversion of RNA to
cDNA using reverse transcriptase (RT-PCR), determination of gene expression of
both receptors with the method Real Time PCR and evaluation of the results.
The results were negative for all samples, which probably means that none
of the two receptors are not expressed at the stages of 2-, 4- and 8-cells.
This may be related to the fact that the embryos in these stages have not
entered yet into the uterine cavity and therefore does not require the
expression of the CRH hormone.
