Summary:
Matrix Solid Phase Dispersion was applied to the effective and quantitative
extraction of Bisphenol A (4,4'-(propane-2,2-diyl)diphenol) from human breast
milk. A reversed phase high performance liquid chromatographic method was
further developed and validated. The mobile phase consisted of CH3CN-H2O (70:30
v/v) and was delivered isocratic ally. The analytical column was Hichrom
Kromasil 100-5C18 (250 4.6 mm) 5 μm (Hichrom, Reading, Berkshire, UK) and was
operated at ambient temperature. UV detection was performed at 230 nm. Sample
preparation was achieved with matrix solid phase dispersion (MSPD) using the
sorbent of Lichrolut cartridges with high recoveries (95.5%). The detection
limit of the method was 5.3 μg/kg. Triethylene glycol dimethacrylate (TEGDMA)
was used as internal standard. Validation of the method was performed in terms
of accuracy and precision: within-day assay (n=6) and between-day assay (n=
33) and was found to be satisfactory, with high accuracy and precision
results. The developed method was comparatively applied to the analysis of
commercial cow’s milk samples with success.
