Summary:
The list of prohibited substances, which is renewed and announced annually from
the World-Anti Doping Agency, includes a variety of low molecular weight
substances. These substances are varied in structure, physicochemical
properties and pharmacology’s behavior (e.g. anabolic agents, stimulants,
narcotics, diuretics, corticostereoids). The aim of this study was to develop
novel methods for the confirmation procedure of these substances. This process
leads to avoid the common but time-consuming stage of their preparation. The
developed methods are specific for each substance and are based on their
different characteristics.
LC-ESI/QTOF-MS methodology included a) the optimization of the ion source
parameters to obtain the maximum abundance of the protonated molecular ion of
each analyte b) the optimization of the in source CID fragmentation and
off-source Q2 fragmentation parameters so as to obtain three diagnostic
fragments for each analyte along with the protonated molecular ion. The
identification parameters that were evaluated are the retention time, the
molecular ion, the fragmentor voltage, collision energy and the number of ion
fragments. The method was applied in multi-component solutions which were
prepared in the laboratory.
