Summary:
The object of this work under the master's degree is divided into two parts. In
the first part isolated with suitable extraction methods, total polar lipids
from tissue of farmed enriched bream fillet fish (Sparus aurata). Then the
total polar lipids used to enrich standard rabbit food and then we conducting
in vivo experiment. In the in vivo experiment, we wanted to study the
cardioprotective properties of polar lipids of enriched bream. The results
showed significant reduction of atherosclerotic plaques in rabbits following
the diet enriched with lipids seabream compared to those which were fed with
diet control. The second part is the isolation of total lipids of farmed Sea
Bass (Dicentrarchus labrax) by the appropriate extraction methods and their
separation to total polar and total neutral lipid fractions. Subsequently, the
total polar lipid fractions were further separated by reversal-phase
High-performance Liquid Chromatography (HPLC) and then were tested in vitro for
their ability to either induce platelet aggregation or inhibit the PAF induced
washed rabbit platelet aggregation, who is considered to be the main cause of
atherogenesis, in order to specify compounds with interesting biological
activity. The fractions which showed the strongest aggregation and inhibition
were sent for identification of their structure with LC-MS/MS.
Electrospray-mass spectrometry elucidated that the lipid composition of the two
fractions contained various diacyl-glycerophospholipids species, where the
majority of them have either 18:0 or 18:1 fatty acids in the sn-1 position and
either 22:6 or 20:2 fatty acids in the sn-2 position for MS1 and MS2
respectively.
