Summary:
In this diploma thesis took place a targeted isolation and structural
identification of coumarinic derivatives from plant material of Micromelum
falcatum Tan. Rutaceae. This plant was selected from five pharmaceutical plants
of the Rutaceae and the Apocynaceae families which were collected in Cambodia.
The set of the plants was extracted in a small scale and were submitted in a
first control in order to define their phytochemical profiling. All the
resulted extracts were evaluated qualitatively using a novel HPLC-DAD method.
The analytical profiling revealed the presence of coumarins as the major class
of constituents in Micromelum falcatum extracts. The HPLC-DAD method was
transferred to semi-preparative scale and was used for the isolation of the
detected coumarins. Eight coumarins were isolated from three different extracts
and three among them consisted new natural products (Microcoumaririn,
Microfalcrin and Micromelosidester) and two others were isolated for the first
time from Micromelum falcatum (Micromarin A and Micromarin B). The
identification of the isolated compounds was performed by HRMS and NMR (1 &2D)
spectroscopy. Secondly, a method LC-HMRS & HRMS/MS was developed in order to
detect and characterize the isolated coumarins in all the extracts as well as
to detect other coumarinic derivatives with similar fasmatometric
characteristics. Furtheromore, two bioassays were performed, in order to
estimate the anti-inflammatory properties of two selected coumarins,
Microminutin (with a lactonic ring) and Micromarin B (with a lateral chain) as
well as EtOAc extracts’.
