Summary:
The present study evaluated the action of the thermophilic lipase of a
Geobacillus strain in non-conventional systems. The term “non-conventional
systems” refers to: 1) the adjustment of High Hydrostatic Pressure HHP
(1,000-10,000 atm), 2) the use of Organic Solvents, in order to stabilize or
even increase the enzyme activity. Primarily, the enzyme purification required
the use of chromatography techniques (the enzyme had been already overexpressed
by the baculovirus flashBACTM system). The enzyme purity was then checked with
SDS-PAGE. The hydrophobic nature of lipase led to the enzyme immobilization and
to its further exploitation in this form. Esterification and
trans-esterification reactions took place in order to evaluate the enzyme in
applications of biotechnological interest. The results proved that lipase
activity increased with HHP application at 3,000 atm as also with enzyme
stabilization in organic solvents.
