Summary:
The enzyme L-Dopa decarboxylase (Ddc) catalyses the decarboxylation of L-Dopa
to dopamine and 5΄hydroxy-tryptophan to serotonin and is found in all aminergic
cells in the CNS. In the present project, Ddc from human and alternative
isoform from rat Ddc-4 expression and parameters of regulation are studied. For
that reason, transfection of the above mentioned cDNAs into CHO cell line,
which do not express Ddc, is carried out. Also, SH-SY5Y cell line is used,
which is a model of neural function, for study in CNS. For the regulation of
the enzymes’ activity, agents that affect the PK-C pathway are used:
Staurosporine (STS) that is an inhibitor of PK-C and PMA that is an inducer of
it. Results of the project showed that in both cell lines, SH-SY5Y and CHO/Ddc,
STS resulted in an increase in both expression and enzymatic activity of Ddc,
whereas in cell line CHO/Ddc-4 there wasn’t any notable change. The incubation
of PMA didn’t have any significant effect on either the expression or the
activity of Ddc isoenzymes. Further investigation of the phenomena observed
here will result in a better understanding of the expression and the enzymatic
activity’s regulation of this complex mammalian enzyme in CNS.
