Summary:
In this study the development of identification and quantification methods for
polyphenols is presented. A method for the extraction and analysis of
hamamelitannin in a pharmaceutical ointment using reversed-phase liquid
chromatography is developed. In subsequent experiments, the objective was to
develop, using the polyphenolic substances gallic acid, ethyl gallate, caffeic
acid, 2-(4-hydroxyphenyl)ethanol and (-)-epicatechin in a standard mixture, a
method for the separation and detection of polyphenols by two-dimensional
high-performance liquid chromatography. After optimization of the operational
conditions, the method was subsequently applied to wine and tea samples, using
on-line two-dimensional liquid chromatography with reversed-phase separation
mode in both dimensions. Regarding the project with the pharmaceutical
ointment, various extraction methods and suggestions for further research are
presented. Concerning the two-dimensional separation, it is proved that even
the combination of two chromatographic columns with a low degree of
orthogonality in their separation mechanisms is a suitable option in the
analysis of polyphenols.
