Comparison of drugs dissolution profiles: Evaluation using computational simulations
Summary:
The process of dissolution is one of the most important processes in many
cases determine the final absorption of the drug. Thus the indicators
characterizing and comparing the dissolution two formulations in vitro are of
great importance. In the current thesis, the drug dissolution profiles:
immediate and modified release were studied and compared using computational
simulations. The similarity of dissolution curves was evaluated via in vitro
dissolution factors, similarity factor(f2) and difference factor(f1).
Moreover, the sensitivity of the aforementioned factors was assessed regarding
the dissolution rate of the drug. More specifically, in order to investigate
and compare the behavior of f1 and f2 factors, our study was performed in two
main levels:
A)'analytical evaluation "of equations f1 and f2. Afterwards the behavior of a
factor was investigated in relation to the other for a number of different
forms of the dissolution curves.
B) Using computer simulations. In that case, the dissolution data for the two
formulations (test and control) was regenerated assuming a set of fast and slow
dissolution rates. The factors f1 and f2 were applied to the same data, and
thus the direct comparison became possible.
