The aim of this study is the synthesis and characterization of novel
mixed-ligand complexes fac-[M(CO)3(L1,2)(P)] and M(CO)2(L1,2)(P)2 were M = Re
or 99mTc, P = PPh3 and L1Η is picolinic acid, a NO bidentade monoanic ligand as
well as the development of an easy method for conjugating a biological vector
in a NO bidentade ligand. The key-compound for this synthesis is
2,3-pyrazinedicarboxylic anhydride. By reacting anhydride with
1-(2-methoxyphenyl)piperazine , L2H is easily and efficiently prepared (95%).
L2H acts as NO bidentate ligand. Rhenium complexes were characterized by IR,
NMR and elemental analysis. The structure was elucidated by X-ray
crystallography. The preparation of 99mTc complexes was obtained by using
fac-[99mTc(H2O)3(CO)3]+. Addition of the NO bidentade ligand to the solution
resulted the fac-[99mTc(CO)3(L1,2)(Η2Ο)] complexes in high radiochemical yield.
Addition of PPh¬3 in the reaction mixture yielded two products,
fac-[99mTc(CO)3(L1,2)(P)], and 99mTc(CO)2L1,2(P)2. 99mTc complexes were
characterized by HPLC, using the corresponding rhenium complexes as references.
All complexes were stable against histidine and cysteine challenge. It can be
concluded that [Μ(CO)3(L1)PPh3] and [Μ(CO)2(L1)PPh3)2] can be used as suitable
building blocks. Biodistribution studies of fac-[99mTc(L2)(P)(CO)3], and
99mTc(L2)(P)2(CO)2 in mice showed that the complexes are stable and have the
ability to cross slightly the intact BBB
