Summary:
Human cytomegalovirus (HCMV) is a DNA virus that is not life-threatening in
normal population, however shows significant mortality and morbidity at
high-risk groups, like infants or immune suppressed patients. Current antiviral
drugs are mostly nucleoside analogues and even if they are effective even
against wild type and mutant forms of the virus, they have quite serious side
effects which in turn do not allow long term treatment. Some nucleoside
derivatives of benzimidazole have also been tested, that possess improved
pharmacokinetics and reduced side-effects. This has prompted us to design and
synthesize a number of new structural analogues which are nucleosides of
imidazopyridine, in order to study the biological importance of the additional
nitrogen atom to the potential anti-HCMV activity of the compounds. We thus
present in this thesis the synthesis of four new nucleosides, the 1- and
3-position isomers of imidazo[4,5-b]pyridine, as well as the corresponding
isomers of imidazo[4,5-b]pyridine-2-one. The synthetic procedures have been
optimized and all new compounds have been adequately characterized using 1D and
2D-NMR techniques and mass spectrometry.
