Summary:
The plant Strychnos aff. dariensis (Loganiaceae) has been used in the past for
the preparation of poisons arrows with curare from the South American Indian
hunters and is used until today from the tribe Yanesha as a medicine. To the
best of our knowledge this work is the first report for the phytochemical
investigation of this plant. To this end, the collected stem bark of Strychnos
aff. dariensis was evaluated for the best extraction procedure in order to
recover a vast range of metabolites. Profiling of the extracts with TLC
revealed that the best approach was the maceration of the stem bark with
EtOAc-EtOH-NH3 (96:3:1) and then extraction with EtOAc and MeOH consecutive.
The isolation procedure was performed using chromatography techniques (TLC,
MPLC, HPLC, FCPC, Sephadex LH-20). Structure determination was based on 1D, 2D
NMR experiments and ESI-HRMS techniques. In total 12 metabolites were isolated,
that can be divided in four groups: phenolic acids, flavonoids, lignans and
alkaloids. Finally, it is interesting to point out that in species of Strychnos
with a relatively low percentage of alkaloids, 11-methoxy-diaboline constitutes
the main component, something that was observed also in Strychnos aff.
dariensis that was investigated.
