Summary:
This thesis focuses on the phytochemical study of the plant Echinops
spinosissimus. Interest for the specific plant grew after its study among 20
greek plants regarding their agrochemical properties. Thε preliminary study
took place during AgroCos, a european research project.
Echinops spinosissimus s a spiny plant, usually found in arid lands near the
sea and especially in the Aegean islands. Extracts and substances from the
plants of the genus Echinops have shown hepatoprotective, antifungal,
antibacterial and insecticidal activity.
Main objective of this thesis is the phytochemical evaluation of Echinops
spinosissimus in order to identify the causes of the insecticidal activity that
it presented during the preliminary study. The ethyl acetate extract was
submitted to fast centrifugal partition chromatography (FCPC) and then
secondary metabolites were isolated using low pressure liquid chromatography
(LPCC), medium pressure liquid chromatography (MPLC) and preparative thin layer
chromatography (prep-TLC). The structura elucidation of the isolated
metabolites was based on experiments with nuclear magnetic resonanse (NMR) and
gas chromatography-mass spectometry (GC-MS).
In conclusion, 10 triterpenes were isolated from the ethyl acetate exctract [3β-
acetoxy-12-oleanen-11-one (1), 21-hydroxy-taraxen-3-acetate (2), 3β-
acetoxy-taraxaster-20-en-30-aldehyde (3), stigmast-5-en-3β-ol (β-sitosterol)
(4), 3-hydroxy-lup-20-(29)-en-30-al (5), 3β-hydroxytaraxaster-20-en-30-al (6),
20-oxo-30-nortaraxast-21-en-3β-ol (7), lup-20-en-3β-ol (lupeol) (8),
5,22-stigmastadien-3β-ol (stigmasterol) (9), 3β-acetoxy-30-norlupan-20-one
(10)].
