Summary:
Insulin-like growth factor (IGF-1) seems to play an important role in the development of uterine leiomyomas. Despite the progress that in elucidating the risk factors, that facilitate leiomyomas appearance and growth, our knowledge with regards to signaling pathways been envolved remains scarce. The elucidation of the molecules that participate in the signaling pathways of leiomyomas appearance, is an urgent need for the long-term therapy of uterine leiomyomas, which does not exist. A better study of IGF-1 isoforms in uterine leiomyomas, can guide us to better understanding of the disease which may be utilized for the searching of new diagnostic and therapeutic tools. In the present study, we evaluated IGF-1 isoforms and IGF-1R expression (IGF-1E1a, IGF-1Eb, IGF-1Ec, IGF-1R respectively) in leiomyomas and adjacent myometrium. Tissue samples were collected from 45 patients who underwent hysterectomy for the treatment of symptomatic leiomyomas. Patients were stratified according to menstrual cycle phase (proliferative phase, secretory phase, postmenopausal patients). mRNA analysis of the three isoforms and IGF-1R, showed that are expressed in both leiomyomas and myometrium οf the patients, with differing expression patterns. In total samples, there was significantly important overexpression of IGF-1Ea isoform and IGF-1R receptor in fibroids compared to myometrium. Also, in women being in menopause, IGF-1Ec levels in fibroids were overexpressed compared to myometrium. Comparing the different phase cycles between them, there was no significantly important difference. These expression patterns could help us develop targeted clinical protocols for the diagnosis and treatment of leiomyomas.
