Summary:
The purpose of this study was to develop a new method for the quantitative measurement of oleocanthal and oleacein, the two major conjugated phenols in extra virgin olive oil, with the use of a spectrophotometer and a specific colorimetric reaction. The reason for the development of this method is the importance of these two components of olive oil, regarding its health protective properties. After the development of the new method, it was compared with two established methods of phenols measurement; the NMR and the Folin-Ciocalteu method.
In the first part of this study, the botanical characteristics of Oleaceae family and the characteristics of olive varieties in Greece were presented. Then the method of production, the categories, the quality and authenticity criteria of olive oil were reported. Also the chemical classes and biological properties of the olive oil components are summarized. Finally, the methods of quantitative and qualitative measurement of phenols in olive oil and most specifically the principle on which the method was based and which was used in this study were reported.
In the second part, the development of the new methods for the optical measurement of the index D1 (the sum of oleocanthal and oleacein) and the quantitative measurement of D1 using a spectrophotometer is described. Moreover the ways of isolating conjugated phenols such as oleocanthal, oleacein and the aldehyde form of the aglycon of oleuropein are reported in detail. Also the methods NMR and Folin- Ciocalteu which were compared with new method are also analysed. An important part of this work involves the application of the method of direct photometric quantification of the D1 index to a big number of Greek olive oil samples and its comparison with the other two methods.
Finally, in collaboration with the science laboratory of Democritus institute, a device, called ARISTOMETRO™, was developed to calculate more easily and quickly the sum of oleocanthal and oleacein by measuring the absorption at a particular wavelength .
