Summary:
In the present thesis the ligand 2, 9-bis(hydroxymethyl)-1,10-phenanthroline, which is a derivative of 1,10-phenanthroline, was synthesized. The corresponding Cu(II) complex was successfully synthesized and fully characterized by various spectroscopic methods (IR, NMR, MS, UV-Vis and fluorescence).
The selection of the ligand and the metal center was made in order to exploit the selective anti-tumor activity of the 1,10- phenanthroline molecule and the cytotoxic activity of the Cu (II) metallic center, aiming at synergistic efficacy.
In addition, their binding to CT-DNA has also been evaluated by UV-Vis spectroscopy, fluorescence, circular dichroism and viscometry. The data reveal that both the ligand and the complex strongly interact with CT-DNA, each one in different ways. The ligand interacts stabilizing the B- structure of DNA. On the other hand the most possible way of interaction of the Cu (II) complex areis the classic intercalation. Preliminary biological studies show that the ligand, PDALC and the complex Cu(II)-PDALC don’t present significant cytotoxicity on tumor cell lines of human osteoblastoma (MG 63 Series). The presence of the Cu(II), however increases the toxicity.
