Summary:
RJ is a thick, milky secretion of hypopharengeal and mandibular glands of worker bees Apis mellifera and it is a sole food of queen bee during her larval and adult life. RJ has been demonstrated to possess several pharmacological activities such as antimicrobial, antioxidant, anti-inflammatory and anti-hypercholesterolemic due to its unique fatty acids content. Also it consists of proteins, sugars, vitamins and amino acids.
The subject of the present Master thesis is the characterization of the quality of royal jelly (RJ) samples from the Greek market by mainly GC-MS techniques. 24 samples were analyzed qualitatively and quantitatively, among which one sample from Crete was further chemically analyzed, for the first time to our knowledge, by chromatographic techniques in order to isolate and determine through modern spectral means, the structures of its secondary metabolites.
The dichloromethane (DCM), methanol (MeOH) and water (H2O) extracts were subjected to various chromatographic separations and 10 metabolites were isolated and structurally determined by modern spectral means. Five fatty acids (octadecanoic acid, trans 10-acetoxy-2-decenoic acid, 10-acetoxydecanoic acid, 10-hydroxydecanoic acid and trans 10-hydroxy-2-decenoic acid) along with one aliphatic ketone (9-hydroxy-2-nonanone) and an aromatic compound (p-hydroxybenzoic acid methyl ester) were isolated from DCM extract, while from methanol (MeOH) one sterol (24-methylenecholesterol) and a fatty acid (9-hydroxy-2-decenoic acid) were isolated. One fatty acid (3,10-dihydroxydecanoic acid) was isolated from water extract. All isolated secondary metabolites, have been previously referred and determined in royal jelly cited in previous studies, according to the international literature. It is noteworthy that four compounds among them (para-hydroxybenzoic acid methyl ester, 10-acetoxydecanoic acid, trans 10-acetoxy-2-decenoic acid and 9-hydroxy-2-nonanone) have been determined by GC-MS and isolated for first time by our lab team in 2005 from a greek royal jelly sample (Arta).

Concerning its biological activities, royal jelly, was tested for its the antimicrobial and antioxidant activity of 10-hydroxy-2-decenoic acid in vitro before and after its encapsulation in liposomes. Moreover, the effect of a greek sample (Arta) in nervous system in an in vivo study in Wistar male rats (Maze water test) was used to estimate the spatial memory levels and the concentration of monoamines and neurotransmitters at specific brain regions.
