Summary:
Ropinirole is a drug approved for the treatment of Parkinson’s disease symptoms and it belongs to the non-ergot dopaminergic agonists. The dopaminergic agonists activate the dopaminergic receptors in the brain and are thus mimicking the effects of dopamine in the brain. Ropinirole possesses affinity for the family of D2 dopaminergic receptors. In the present work, a method for the quantification of ropinirole and its impurities in tablets was developed and validated, by using a HILIC-ESI/MS approach. The usage of mass spectrometry bestows high sensitivity and selectivity to the method, while simultaneously the hydrophilic interaction chromatography HILIC allows the rapid isocratic elution of the analytes. HILIC chromatography is ideal for performing LC-ESI/MS analysis, since the large percentage of organic modifier in the mobile phase improves the method’s sensitivity. The chromatographic column utilized was ZIC®-HILIC. The mobile phase was composed of 8% ammonium formate buffer 18.8 mM (pH = 6.20) in acetonitrile. The mobile phase flow rate was 0.25 mL min-1. For the ion scanning, the single ion monitoring (SIM) technique was selected. The time of chromatographic analysis does not exceed 15 minutes. The developed method was successfully applied to the analysis of ropinirole tablets.
