Summary:
Prader-Willi syndrome (PWS) is a relatively common genetic disorder. About ¼ of adult patients with the syndrome, suffer from type 2 diabetes mellitus (DM) and obesity. Due to existence of several comorbidities and the complex pathophysiology of the syndrome, appropriate DM treatment in these patients requires further verification. A systematic review of trials in adults with PWS that include patients with DM or study glucose metabolism factors, has been performed for this purpose. Only very few trials have been found, concerning mostly growth hormone treatment or exenatide administration. The majority of them was of short duration with a limited number of participants. As a conclusion, current evidence does not support the utilization of different medical treatment for DM in patients with PWS in comparison to DM patients without the syndrome. However, due to multiple confounding factors, further larger multicentre trials need to be organized to study possible effects of current and new treatment options for PWS on glucose metabolism.
