Summary:
In this study, we examined the production and concentration of various biochemical factors in the amniotic fluid of the second trimester in pregnancies with trisomy 13 and 18 and compared them with measurements in the amniotic fluid of normal karyotype embryos.
We chose to study the amniotic fluid because it reflects the extracellular environment of the fetus. Amniocentesis was performed during the first half of the second trimester of pregnancy for prenatal diagnosis. In particular, leptin, resistin and TNF-α content was studied in amniotic fluid of trisomy 13 and 18 and normal karyotype pregnancies.
Leptin is a hormone secreted by adipose tissue and associated with food intake. It was measured in 3 amniotic fluid samples with trisomy 13, in 11 cases with trisomy 18 and 23 amniotic fluid samples with normal karyotype. The results showed that there is no statistical significance.
Resistin is a hormone that is also secreted by adipose tissue and appears to play a key role in inflammation. It was measured in 3 trisomy samples 13, 11 samples with trisomy 18 and 23 samples of amniotic fluid with normal karyotype. The mean concentration of resistin in normal karyotype embryos is statistically significantly higher than the mean concentration in trisomy 18 fetuses.
The last measured mediator, TNF-α (Tumor Necrosis Factor alpha), plays an important role in the immune system, insulin resistance, and exerts significant anti-inflammatory activity. It was also measured in 3 amniotic fluid samples with trisomy 13, 11 samples with trisomy 18 and 31 samples with normal karyotype. From the comparison of TNF-α values with amniotic fluid values with normal karyotype, it was found that the mean concentration of TNF-α in normal karyotype embryos was statistically significantly higher in relation to the mean TNF-α concentration in trisomy 18. These data indicate a possible involvement of TNF-α in metabolism οφ embryos with abnormal karyotype.
