Summary:
STAT5 is a transcription factor than can affect gene expression by interacting with other proteins such as LSD1 and HDAC3. Using Cytoscape, lists of confirmed protein interactions for STAT5a, STAT5b, LSD1 and HDAC3 were analysed and found to have plenty of common interacting proteins. Among them EZH2 appeared important as it interacts with all four of the aforementioned proteins. With the help of GeneCodis, mass spectrometry lists from Ba/F3 cells’ cytoplasmic and nuclear extracts, where STAT5

complexes were isolated by in vivo biotinylation, were analysed. The analysis confirmed cellular pathways involving STAT5. Also, the use of an LSD1 inhibitor on Ba/F3 cells showed, via RT-PCR, that the inhibition of LSD1 results in the overexpression of the Gfi1b gene, which is a common target gene of LSD1 and STAT5a. Thus, this inhibitor is functional in Ba/F3 cells and could be used in future experiments. Finally, immunoprecipitation experiments were performed to validate the interaction between LSD1 and CNOT2. This work highlighted proteins that possibly interact with STAT5 and cellular pathways where STAT5 participated. Also, it confirmed the functionality of an LSD1 inhibitor, which could be used in the future in therapeutic approaches for diseases linked to STAT5 deregulation.
