Summary:
STATs (Signal Transducers and Activators of Transcription) play a significant role in normal cellular functions, as well as in diseases, including solid tumors and hematological malignancies. Among STAT family members, STAT3 and STAT5 are mainly implicated in leukemias and thus constitute interesting therapeutic targets.
In order to study the role of STAT5 and STAT3 in acute myeloid leukemia (AML), in the present study, bioinformatics analysis of RNA-seq data was performed. The data were obtained from wild type and knock-down (KD) MOLM-13 cells, which were transduced with lentiviruses encoding shRNAs to KD either STAT5A or STAT5B. Differentially expressed genes were identified in the STAT5A and STAT5B KDs, respectively. In the case of STAT5B, the biological pathways, that were affected, were also identified in an attempt to understand the mechanism by which STAT5B is involved in the pathobiology of AML. In addition, MOLM-13 and Kasumi-1 cells (AML cell lines) were also transduced with lentiviruses encoding shRNAs to KD STAT3. KDs were confirmed by RT-qPCR and Western Blot and will be used, in the future, for RNA-seq experiments and bioinformatics analyses.
