Summary:
The neoplastic disease is the second cause of death worldwide with established treatment of surgery, chemotherapy, radiation and immunotherapy. However side effects and the reduction of quality of life are common. Basidiomycete Lentinus Edodes is the second most famous edible fungus worldwide and has been studied due to its antioxidant and antineoplastic properties. Since 1970, agents such as polysaccharides (Lentinan, LEM, LEP, LEP1) , proteins (Latcipin-1, Latcripin-3, Latcripin-11, Latcripin-13) and proteoglycans (LE-MGG, LAP-1) have been isolated and studied. Their individual administration and their combination with chemotherapeutic agents (Oxaliplatin, Cisplatin, S-1) were studied. The mechanism of the agents derived from the fungus Lentinus Edodes has been described extensively. Lentinan was found to cause an immune response, where the activation of phagocytes leads to the excitation of macrophages - due to increased cytokine production - resulting in the activation of TH-1 cells and adaptive immunity (T and B cells). Polysaccharides and certain proteins were found to cause apoptosis, which depends on Caspase-3, Caspase-9, increasing the expression of the Bax gene and reducing the expression of the Bcl-2 gene. Apoptosis is due to both exogenous and endogenous signaling route and confirmed by morphological changes in neoplastic cells. Lentinan is also acting through angiogenesis suppression, which depends on T-cells and confirmed by reduced expression of the vascular protein agent (VEGF). Latcripin-1 protein was found to cause autophagy, which is regulated by the PI3K/AKT/mTOR signaling pathway. The agents that were isolated only affect neoplastic cells, with no reported adverse reactions, however further research are needed to establish them as an antineoplastic medicinal product.
