          <dc:description xml:lang="el">Ο ινσουλινομιμητικός αυξητικός παράγοντας (IGF-1) αποτελεί σημαντικό  βασικό ρυθμιστή κυτταρικών διεργασιών όπως ο πολλαπλασιασμός, η  επιβίωση και η απόκριση σε στρες. Από την πρόδρομη μορφή του (proIGF-1)  μπορούν να προκύψουν τρεις ισομορφές μέσω εναλλακτικού ματίσματος. Ο  IGF-1Ea που είναι η πιο άφθονη από τις τρεις, και οι IGF-1Eb και IGF-1Ec  που φυσιολογικά υπάρχουν σε πολύ μικρή ποσότητα στα κύτταρα. Το  πεπτίδιο Eb της ισομορφής IGF-1Eb έχει συνδεθεί με την απόκριση σε  στρεσογόνους παράγοντες και προηγούμενες παρατηρήσεις μας έχουν δείξει  πως εισέρχεται στον πυρήνα μετά από έκθεση σε UV ακτινοβολία, γεγονός  που υποστηρίζει τον ρόλο του στην απόκριση σε στρες.    Στην παρούσα διπλωματική εργασία μελετήθηκε η επίπτωση της  αποσιώπησης της ισομορφής IGF-1 Eb στη φυσιολογία του κυττάρου. Για τον  σκοπό αυτό πραγματοποιήθηκε knockdown με siRNA σε τρεις κυτταρικές  σειρές (PC3, MDA-MB-231 και HepG2), με στόχο την σύγκριση του  πολλαπλασιασμού, της βιωσιμότητας, της απόπτωσης και του κυτταρικού  κύκλου, ανάμεσα στην ομάδα ελέγχου (WT) και σε knockdown (EbKD)  κύτταρα, με και χωρίς την έκθεση σε UV ακτινοβολία ως παράγοντα στρες.  Επιπλέον, χορηγήθηκε εξωγενώς πεπτίδιο Eb τόσο σε WT όσο και σε EbKD  κύτταρα που υποβλήθηκαν σε UV στρες, ως συμπληρωματική αξιολόγηση  της επίδρασής του.  Τα αποτελέσματα υποδεικνύουν ότι η απουσία Eb προκαλεί διαφοροποιήσεις  στον πολλαπλασιασμό των κυττάρων, ακόμη και απουσία στρες, με  διαφοροποιήσεις ανά κυτταρική σειρά. Μετά από UV στρες τα EbKD κύτταρα  φάνηκαν πιο ευάλωτα, υποδεικνύοντας ότι το πεπτίδιο Eb διαδραματίζει ρόλο  στην απόκριση στο στρες.</dc:description>
          <dc:description xml:lang="en">Ιnsulin-like growth factor 1 (IGF-1) is a key regulator of key cellular processes,  including proliferation, survival, and stress responses. Three IGF-1 isoforms  can be generated from its precursor form (proIGF-1) through alternative  splicing: IGF-1Ea, which is the most abundant isoform, and IGF-1Eb and IGF 1Ec, which are normally expressed at very low levels in cells. The Eb peptide  derived from the IGF-1Eb isoform has been associated with cellular  responses to stress, and our previous observations have shown that it  translocates to the nucleus following UV irradiation, supporting its role in  stress response mechanisms.  In the present thesis, the impact of IGF-1Eb isoform silencing on cellular  physiology was investigated. To achieve this, siRNA-mediated knockdown  was performed in three cell lines (PC3, MDA-MB-231, and HepG2), aiming to  compare cell proliferation, viability, apoptosis, and cell cycle distribution  between control (WT) and knockdown (EbKD) cells, both in the presence and  absence of UV irradiation as a stress-inducing factor. In addition, exogenous  Eb peptide was administered to both WT and EbKD cells subjected to UV  stress, as a complementary assessment of its functional role.  The results indicate that the absence of the Eb peptide leads to alterations in  cell proliferation even under non-stress conditions, with differences observed  among the examined cell lines. Following UV exposure, EbKD cells appeared  more vulnerable, suggesting that the Eb peptide plays an important role in the  cellular stress response.</dc:description>
