The role of molecular markers nestin and cd146 (mcam) in breast cancer based on the biological mechanisms of action of the tumor stem cells
Summary:
Background&Purpose: To understand the expression of CD146 (MCAM) and nestin in
different molecular subtypes in breast cancer.
Subjects&Methods:141 cases including paraffin-embedded tumor tissues were
selected in 2000-2014, classified into luminal A, luminal B, HER2+ and
triple-negative subtypes. Expression of MCAM and nestin were analyzed using
immunohistochemistry.
Results: Nestin was highly expressed in triple-negative tumors (p <0,001), but
not in patients exhibiting the HER2+, luminal B and luminal A subtype. MCAM was
highly expressed in the triple-negative tumors (p <0,001) and decreasingly
expressed in the HER2+, luminal B and luminal A subtypes. Expression of both
nestin (p = 0.0021) and MCAM (p = 0.008) were statistically related to the
histological grade and the Ki-67 cell proliferation marker (p=0,007 and p<0,001
respectively). MCAM expression was associated with the disease stage (p =
0.006), whereas nestin expression was statistically related to the younger age
of the patients (p = 0.029). Nestin and CD146 affect the occurrence of relapse
time (p = 0.022 and p = 0,003 respectively) while CD146 is an independent
prognostic factor for relapse time (p = 0,037) in breast cancer.
Conclusions: Nestin and MCAM expression were associated with aggressive tumor
features and clinical behavior of breast carcinomas exhibiting poor prognosis.
