Summary:
In this thesis, the effect of diabetes on the musculoskeletal system is
studied. Particularly, the object of the study is to evaluate the effect of
metabolic and endocrine changes due to diabetes on bone quality and degradation
of the Achilles tendon. The mechanical response of the tibia and the Achilles
tendon and the correlation of possible pathological events with diabetes are
being investigated. Imbalance of expression of matrix metalloproteinases and
their inhibitors, contribute to abnormal connective tissue degradation. In
orthopaedics, their clinical applications are constantly being explored. With
these data in this PhD study the role of metalloproteinases in poor quality
bone and the degradation of the Achilles tendon on diabetes are also evaluated.
To perform the experiment 40 male rats (Wistar rats) were used. Diabetes was
induced by administration preoperatively Streptozotocin (STZ 55 mg / kgr
weight). After the installing of diabetes, a euthanasia of animals and
isolation of the Achilles tendon and the tibia were performed in order to carry
out biomechanical evaluation. In a parallel manner MMP-2 levels in blood, MMP-2
and 9 Achilles tendon, was measured as well as the analysis and evaluation of
all results and histological confirmation.
It was found that diabetes affects the musculoskeletal system: as it affects
the mechanical properties of bones and the Achilles tendon. Metalloproteinases
are associated with the progressive deterioration of the quality of the bone of
the tibia, and also pathological processes in the Achilles tendon of diabetic
rats. The mechanical differences are connected with biochemical changes in
diabetic Achilles tendon and the tibia.
