Summary:
In the present study, we conducted a clinical evaluation of the mRNA levels of
the genes DDC and BCL2L12 and their corresponding regulatory microRNAs, miR-145
and miR-182, in surgically removed renal tissue specimens and cultured renal
cancer cells exposed to the anticancer drugs gemcitabine, sunitinib, and
everolimus. For that purpose, 4 distinct quantitative real-time PCR protocols
were developed and sequentially our data were subjected to statistical
analysis. According to our results, DDC was downregulated in malignant samples
and correlated with the histological subtype and the grade of malignancy of
cancerous samples. Regarding BCL2L12, this likewise demonstrated downregulation
in malignant renal tissues, but no statistically significant correlation with
the clinicopathological parameters. Moving to miR-145, its levels also
demonstrated downregulation in tumor samples, as well as correlation with the
histological subtype and the stage of the disease. As for our cell culture
experiments, significant alterations were detected only in the case of miR-145
and specifically with regard to the cancer cells exposed to sunitinib and
everolimus. In conclusion, the completion of the present study revealed that
the mRNA levels of DDC and BCL2L12, as well as the RNA levels of miR-145 could
be considered as promising molecular markers for kidney cancer.
