Summary:
The present Master Thesis is about the evaluation of a quantification and a
confirmation method in athletes urine using gas chromatography – mass
spectrometry of the following endogenous anabolic steroids: testosterone,
epitestosterone, androsterone etiocholanolone, 5a-androstane-3a,17a-diol,
5b-androstane-3a,17b-diol, testosterone to epitestosterone ratio (T/E) which
compose the athletes .
For the evaluation of the two methods, series of validation experiments were
conducted based on the current International standards for Laboratories, ISL,
of the World Anti - Doping Agency( WADA). The factors that were evaluated
during the validation procedure were the following: chromatographic
characteristics, linearity of the concentration range understudy, sensitivity
and the sensitivity variation, precision, intra-day precision, inter–day
precision, limit of detection( LOD) and limit of quantification (LOQ) and
relative standard combined uncertainty (uc%).
The two methods were demonstrated to be fit –for- purpose and they fulfilled
the specifications of the WADA’s Technical Document TD2014EAAS
