Summary:
Alkannin and shikonin (A/S) are lipophilic red pigments with a wide spectrum of
well-established biological activities, such as wound healing, antimicrobial,
anti-inflammatory, anti-cancer and antioxidant ones. The stability of A/S and
their derivatives during process and storage is crucial for their use as
pharmaceuticals, cosmeceuticals and food additives.
The aim of this diploma thesis was to examine the stability of monomeric
shikonin in several solvents (acetone, methanol, chloroform and n-hexane) under
several conditions (light, temperature and pH). Moreover, the stability of
shikonin in powder form was examined under several storage conditions. The HPLC
system was equipped with a UV-Visible detector, set at 520 nm. The
chromatographic separation was carried out on a C12 Synergi Max-RP column (80A,
4 μm, 150  4.6 mm).
Monomeric shikonin concentration was decreased rapidly in solutions exposed to
sunlight in all solvents and several photodegradation by-products were
detected. The photodegradation of monomeric shikonin confirmed by its exposure
at fluorescent and UV-A irradiation. Shikonin concentration in samples
protected from sunlight was decreased too. Additionally, temperature higher
than 60 oC affected monomeric shikonin stability in methanolic solutions.
Moreover, shikonin in powder form was stable under several storage conditions,
such as sunlight or atmospheric oxygen for more than four months.
