Summary:
Chapter 1 presents data on the disease of type 2 diabetes, the role of insulin
in glucose metabolism and approaches for the treatment of the disease. Then,
there is some information about potential antidiabetic agents and their mode of
action. Moreover, the metabolism of glycogen in the body is described and the
role of its key enzyme of glycogen phosphorylase that catalyses the first step
of glycogen degradation to glucose-1-phosphate. The mechanism of action of the
enzyme is then analysed.
Chapter 2 presents a literature review on glycogen phosphorylase inhibitors
that contain a 2-naphthyl group and a purine group and how these compounds
served as leads for new ligands that were tested experimentally for their
ability to inhibit the enzyme. Then, the aim of the current thesis is
presented, which is the synthesis of organic compounds and the control of their
action as inhibitors of glycogen phosphorylase (GP) in the disease of type 2
diabetes.
In chapter 3, synthesis of a,a-disubstituted aldeydes for the a-amination
reaction is presented and also other synthetic reactions. Furthermore, selected
spectroscopic data of the synthesized compounds are presented.
Chapter 4 describes the experimental part, which includes information about the
isolation of RMGPb from rabbit skeletal muscle. It outlines the experimental
procedure, the ascorbic acid method and the kinetic experiments for the
determination of IC50.
The results of the kinetic experiments are summarized in chapter 5 along with
the IC50 graphs of the compounds that exhibited enzyme inhibition.
In chapter 6 the experimental procedures and the spectroscopic data of all the
compounds are presented.
