Summary:
The human Polybromo-1 protein (PB1) is a unique subunit of the PBAF complex and coordinates key features of chromatin remodeling complexes and chromatin architecture change. Additionally, it is required to progress the cell cycle through mitosis. This protein is an epigenetic target and is used to construct a pharmacophore model.
The aim of this thesis is the heterologous expression and purification of target protein PB1 (5) by using a different purification protocol than that proposed by the SGC (Structural Genomics Consortium) to enable large amounts of protein to be obtained. With protein purification of more than 90%, it is feasible to study the protein with biophysical trials such as Circular Dichroism (CD) and Differential Scanning Fluorimetry (DSF), and to perform preliminary ligand experiments with a protein inhibitor, with a view to finding new candidate drugs targeting this protein.
