IDENTIFICATION AND QUANTIFICATION OF 11-NOR-9-CARBOXY-TETRAHYDROCANNABINOL AND NORPSEUDOEPHEDRINE WITH DIRECT INJECTION BY LC-QTOF/MS
Summary:
The World Anti-doping Association (WADA) renews and announces annualy the List of Prohibited Substances which among other categories includes cannabinoids and stimulants. THCCOOH with a threshold of 150 ng/ml is included in the cannabinoids caregory, while norpseudoephedrine is included in the stimulants category with a threshold of 5μg/ml. In this Master Thesis the development, the optimization, the validation of confirmatory methods for the identification, the quantification and the separation of the above substances in athletes’ urine samples were conducted by direct injection and the use of the principle of liquid chromatography (LC) – mass spectrometry (MS) by quadropole time of flight mass analyzer (QTOF). For reversed phase liquid chromatography was used column Zorbax Eclipse Plus C18 (Agilent Technologies) with a mobile phase consisting of a mixture of water and acetonitrile in various proportions. Analytes were ionized with an electrospray ionization, performed in the positive mode (ESI(+)). The parameters of the identification that were evaluated were the resolution time of the substance, the molecular ion, the voltage of fragmentor source, the collision energy and the number of fragments from the molecular ion. Validation of methods was made according to the current International Standard for Laboratories of the World Anti-Doping Association (WADA) and the ISO 17025. The parameters of validation were the signal to noise ratio, relative retention time, linearity, sensitivity, accuracy, repeatability, intermediate precision, limits of detection and quantification, matrix interferences, specificity, carry over and the combined standard uncertainty. As a conclusion, the method for identification and quantification of THCCOOH and the method of quantification of norpseudoephedrine are considered suitable for use to doping control in athletes’ urine samples.
