Summary:
Alzheimer's disease is the most common progressive neurodegenerative disease, provoking serious mental and emotional complications for the patient. Cholesterol metabolism has been implicated as one of the contributing factors to Alzheimer’s disease, emerge and prognosis. Scavenger Receptor class B type I (SR-BI) is a high density cholesterol (HDL) receptor and mediates cholesterol transport. SR-BI is among others expressed in astrocytes and smooth muscle cells of the brain vessels of Alzheimer's patients while Aβ protein may be a ligand of SR-BI. In order to further investigate the role of the SR-BI receptor of astrocytes in the removal of Aβ deposits from the brain the second transgenic line of mice carrying the human SR-BI (huSR-BI) transgene under control of the GFAP protein promoter (Glial Fibrillary Acidic Protein) was produced. The GFAP promoter is specifically expressed in astrocytes. The production of this second line was performed by microinjection of the GFAP-huSR-BI transgene into fertilized oocytes at the juncture stage which were then transferred to a pseudopregnant female mice.
