Summary:
Silk fibroin (SF) is a protein–based bio macromolecule with excellent biocompatibility and biodegradability, for this reason silk fibroin micro-and nano-particles provide new options for drug delivery. In the present study, aqueous-based preparation methods for silk nanoparticles are presented, based on phase separation between silk fibroin and polyvinyl alcohol (SF/PVA). The encapsulation efficiency in SF/PVA nanoparticles was examined at a range of concentrations from 10 mg/L to 500 mg/L for the following drugs: pramipexole, curcumin and propranolol hydrochloride. The highest encapsulation entrapment for the SF/PVA nanoparticles loaded with curcumin was observed at 500 mg/L, for propranolol at 50 mg/L and for pramipexole at 35 mg/L. The drug release profiles of the above drug molecules were also examined and SF/PVA nanoparticles loaded with propranolol showed the highest release profile. Subsequently, in order to improve the encapsulation efficiency and drug release, a mix of silk fibroin and bovine serum albumin (SF-BSA/PVA) nanoparticles were prepared. Finally, magnetic silk fibroin nanoparticles were prepared targeting their ability to retain and to deliver the therapeutics at the preferred site. SF/PVA micro-and nanoparticles presented to have potential as drug delivery carriers in biomedical applications. The addition of BSA and magnetic nanoparticles improved the drug encapsulation and release.
