Hydrophilic Interaction Liquid Chromatography: Development and Validation of a HILIC-FL method for the determination of octreotide in medicinal products
Summary:
Somatostatin (SS) is a peptide that inhibits the secretion of growth hormone from the pituitary gland. The use of bioactive forms in medical treatment is limited due to their short half-life in vivo. Octreotide is a synthetic, cyclic octapeptide analogue of somatostatin hypothalamic hormone, of which it has a longer half-life and more selectivity in the inhibition of hormone secretion. It is used to control the hypersecretion of hormones in pituitary tumors, endocrine pancreatic tumors and carcinoid tumors. It is indicated for the treatment of acromegaly for the treatment of severe diarrhea and hot flushes observed in neuroendocrine tumors of the pancreas and gastrointestinal tract and for the treatment of esophageal variceal bleeding.
The chromatographic study was performed by hydrophilic interaction liquid chromatography in combination with a fluorimetric detector (HILIC). Chromatographic separation was performed on a Xbridge® HILIC BEH analytical column with isocratic elution and a flow rate of 0,25 mL min-1. The method was validated according to ICH guidelines.
