Summary:
Fluorescein derivatives are widely employed in chemical biology as molecular
probes. Modification of the xanthene skeleton is of great importance, as it can
lead to alterationin physical properties, including emission wavelength, internal
fluorescence quenching and fluorescence quantum yield. In particular,
introduction of strong electronegative substituents on the xanthene ring, like
fluorine atoms, results in derivatives with lower pKa values and singlet oxygen
sensitization quantum yield.
Herein, we describe our attempts to develop an improved synthetic protocol
towards Aarhus Sensor Green, a tetrafluoro-substituted fluorescein analog
bearing an 9,10-diphenylanthtracene moiety. Moreover, we report the synthesis
of a new, non-symmetrical fluorescein analog, bearing two fluorine atoms on
the same ring of the xanthene part of the dye. This product is anticipated to
have similar physical properties with the tetrafluorinated analogs of Oregon
Greens.
