Summary:
In this study Robustness test was performed on a RP-HPLC method that has been developed for rapid simultaneous determination of monoamines neurotransmitters, such as dopamine and its metabolites, norepinephrine, as well as serotonin and its metabolites in quail brain microdialysis samples. The method used for the determination of monoamines focuses on electrochemical detection acquires high sensitivity measurements and can detect these substances at pg levels. Based on the principles of the reverse phase ion-pair liquid chromatography, monoamines were separated on an aquasil C18 column with an isocratic mobile phase consisting of acetonitrile and phosphate buffer at pH = 3. Mobile phase's flow rate was set at 0.2 mL/min and 800 mV were applied during the detection. Robustness test was performed using two level fractional factorial design, so only main effects can be estimated. Factors chosen for this study were % ACN of the mobile phase, phosphate buffer concentration, ion pairing agent concentration (Octyl sulfate sodium salt), concentration of Na2EDTA and the voltage (mV) of the electrochemical detector. The significance of the effects was determined statistically, using the Student (t-test) test, and graphically, through normal and semi-normal probabilities plots and Pareto charts.
