Summary:
Brain cancers, like glioblastoma multiforme (GBM), are practically incurable due to their location and their highly-aggressive nature. The current conventional radiation therapies, such as photodynamic therapy (PDT), which targets the photoactive molecules inside the tumors, have been clinically evaluated, but with non-satisfactory success. This is mainly due to the limited penetration of light into the tissue, the widespread proliferation of cancer cells and the fact that these methods are highly invasive, requiring open surgery.
In this work, the design and synthesis of chemiluminescent-photoactive organic compounds based on luminol are described. N-acyl derivatives of luminol and isoluminol were synthesized and studied. Synthetic routes are based on the acylation of the free amino group of luminol and isoluminol, with chains (of various lengths) bearing phosphonium cations. In addition, an aryl alkyl substituted, mitotropic derivative of luminol was designed and synthesized.
Subsequently, the synthesis and characterization of N-alkylated and N-acylated derivatives of 3-aminophthalic acid is being described, in order to study the mechanism of luminol’s chemiluminescence.
Finally, a fullerene C60 derivative was designed and synthesized to study and evaluate its encapsulation in specially designed nanocages and moreover to study its chemical behavior - activity, being encapsulated, as well as its possible applications.
