Summary:
In this study it was developed Lornoxicam gels for topical administration, thus avoiding side effects from its systemic administration. Topical administration is a suggested route in case of immediate relief and treatment of a local inflammation. Lornoxicam is a non-selective cyclooxygenase 1 and 2 (COX-1, COX-2) inhibitor used to treat pain, inflammation and rheumatoid arthritis. To develop Lornoxicam gel, a Mixture Design was applied with Triethanolamine, Polyethylene Glycol 400 and Water as the components of the mixture and the Saturation Solubility of Lornoxicam in the system as a response. After conducting the experiments, a mathematical model was chosen that best connects the relationship of the three solvents with the saturation solubility of Lornoxicam. The chosen model was the special cubic model with good fit to the experimental data (R2 = 0.9987, R2adj = 0.9974 and R2pred = 0.9788). After all these, the optimization of the response was conducted by requesting from the model to suggest possible solutions for the development of a gel with 1% w / w Lornoxicam. The three formulations had a desirability factor equal to one but saturation solubility experiments were performed to confirm each situation. Finally, these gels were tested in In-vitro percutaneous permeation experiments using Franz-type cells. In this study, the permeability was assessed both through the Strat-M , which is an artificial membrane similar to skin, and through the human stratum corneum.
