Summary:
The development of new potent drugs in oncology consists a rather difficult task, due to the complexity of cancer, the constantly increasing cost of research, and the fact that resistance in therapy is developed in many cases. All these reasons demand the establishment of innovative tools, which would not only enhance our understanding of cancer’s pathophysiology and mechanisms but will furthermore diminish the time and costs of R&D. This is something that can be achieved through utilizing computational approaches in drug development, which can complement well-established laboratory methods and make research overall much more efficient. This systematic review intends to summarize the different in silico approaches which are used in oncology drug development. Structure-based, fragment-based, ligand-based, and integrated methods are some examples, as well as methods that enable drug repositioning. In vitro and in vivo laboratory data can be used in the creation of computational models. These models can make the vast amount of data, which are available today, manageable and yield significant information for the discovery of novel efficient drugs.
