Summary:
The aim of this study is to investigate the suitability and effectiveness of various palynological chemical preparation protocols in the extraction of pollen based on their origin.
For this purpose, different sediment samples of varying composition and origin were collected, while in addition some fresh material from Greek vegetation was included in the study. The chemical preparation procedure that was followed differs significantly between samples as a result of their specific characteristics aiming to retrieve the larger amount of pollen possible from each sample medium. More specifically, in fresh and reference samples, the method of acetolysis was applied, while the method of flotation in heavy liquids was used for the archaeological excavation samples and for the lake and lagoon sediments the classical acid treatment was applied.
The laboratory methods for the extraction of pollen grains were analyzed in this study aiming to improve their extraction in various sediment samples. Finally, some suggestions for improving the pollen extraction methodologies have been done.
