Development and validation of a high-performance liquid chromatography with photodiode array detector (HPLC-DAD) method for the quantitative determination of apixaban and sodium benzoate in oral suspension
Summary:
In this MSc research work, a method was developed and validated for the simultaneous determination of apixaban and sodium benzoate. Apixaban belongs to the category of anticoagulants, preventing the formation of blood clots, while sodium benzoate acts as an antimicrobial preservative.
To this regard a high-performance liquid chromatographic method coupled to photodiode array detection was developed and fully validated. Chromatography was performed on a Symmetry Shield RP18 C18, 150 x 4.6 mm, 3.5 μm analytical column with a mobile phase of sodium acetate solution(pH=4.1): acetonitrile: methanol 65:10:25, v/v/v. The flow rate was set at 1.0 mL/min and the analysis time was less than 25 min. Detection was performed at 230 nm.
The developed method provides an acceptable degree of linearity, accuracy, precision, stability, specificity, and robustness when applied to samples.
