Synthesis and Characterization of Bicomponent Copolymer Poly(ethylene oxide)-b-Poly(glutamic acid) and Coupling it with a Contrast Agent
Summary:
In this thesis, the synthesis of a series of hybrid diblock copolymers of different
compositions. The polymer synthesis was carried out via ring-opening polymerization
(ROP). High vacuum techniques were employed for the synthesis of the Ncarboxyanhydrides, the purification of the solvents, and the final copolymers, ensuring
high purity of the system and absence of impurities that could lead to undesired
reactions. The synthesized copolymers have the ability to self-assemble in aqueous
solutions and form micellar structures on a nanoscale, with the outer hydrophilic shell of
the nanostructures and the hydrophobic core conjugated with the contrast agent drug.
The successful synthesis of the polymers was confirmed using size exclusion
chromatography (SEC), proton nuclear magnetic resonance (1H-NMR), and Fouriertransform infrared spectroscopy (FT-IR). Dynamic light scattering (DLS) was used to
study the self-assembly capability of the copolymers and the size of the synthesized
nanostructures. Additionally, the surface charge of the nanoparticles was studied by
measuring the zeta potential. The study of the properties of these hybrid copolymers is
of particular importance for the development of stable nanostructures that can be used
for targeted and controlled drug release in the body.
