Summary:
Atmospheric organosulfates (OSs) are sulfate esters,present in the atmospheric aerosol. They are a class of organic compounds formed by atmospheric multiphase and heterogeneous chemical reactions and contribute to a significant fraction of the Particulate Matter (PM). OSs are currently under investigation for their ability to induce oxidative stress. The Oxidative Potential (OP) of OSs was determined via the dithiothreitol (DTT) assay. The rate of the reaction, commonly called DTT activity, is determined by measuring the consumption of DTT over time, which is proportional to the concentration of redox-active species in PM extracts. In this study, an in-house method was developed, using 1,4-Naphtoquinone standards, and was applied on PM2.5 aerosol samples, from a domestic area in Nicossia, Cyprus.Briefly, the redox reaction takes place in a mixture of potassium phosphate buffer (pH=7.4), aerosol extractand a known amount of DTT, incubated at 37°C. At designatedtime intervals, DTNB is added to determine the remaining quantity of the DTT. The absorbance of the colored product of the DTT-DTNB reaction is measured at 412nm. DTT activity of the samples presented a seasonal trend, showing higher values during winter time, when the anthropogenic OSs are most abundant. Also, a strong correlation between OP activity and glycolic acid sulfate, an indicator of photo-oxidational processes in the atmospheric aerosol, is observed.
