Summary:
The role of visfatin in oocyte maturation remains unknown yet. This study
investigated the effect of visfatin upon insulin receptor phosphorylation and
this effect depends on the presence of insulin in the culture medium.
Oocytes were used from12 days aged female mice. The oocytes were divided in two
groups and cultured. In the first group insulin was added in the culture
medium, but the second group lacked insulin. Both groups were divided in
subgroups according to visfatin’s concentration used. We used 0 ng(control),
1ng ,10ng, 50ng, 100ng. 12 days later the oocytes were collected, remained in
RNA later in -80°C and were analysed using Western blotting in order to
investigate the insulin receptor phosphorylation.
Visfatin significantly increases receptor’s phosphorylation as the
concentration increases and particularly 50 ng of visfatin makes
phosphorylation to achieve its maximum. However the minimum sign of
phosphorylation is determined to be at 100ng, which appears to give
phosphorylation’s sign lower than 0ng (control).
We conclude that visfatin increases insulin receptor phos.
