Summary:
Multiple Myeloma (MM) remains a major cause of cancer mortality and is considered the second most frequent haematological malignancy in Europe and USA. A limited number of studies have been conducted regarding the PDGF receptors and the correlation with clinical and pathological parameters and prognosis in multiple myeloma. The PDGF receptors are cell surface tyrosine kinase receptors and may play an important role in cancer. The aim of our study was to evaluate, for the first time, the expression of PDGF receptor α and β (PDGFR α and β) in marrow trephine biopsies of MM patients and explore possible correlations with clinical data and angiogenesis as assessed by marrow micro vessel density (MVD). Methods: We studied and assessed retrospectively the relation of expression of PDGFRa, PDGFRβ and MVD in formalin fixed paraffin-embedded bone marrow sections of 120 newly-diagnosed MM patients. The immunoreactivity of PDGFR α and β was examined on the basis of positive plasma cells (PCs) with the following cut off values: <20% positive PCs (negative expression), 20-40% positive PCs (intermediate expression) and >40% positive PCs (high expression). Results: our results identify the positive immunocytochemical (ICC) expression of PDGFR α and β in plasma cells and mainly indicate the correlation of the increased immunoexpression of PDGFR β with advanced features of the disease. The expression of PDGFR α and MDV were not correlated with specific features of the disease. Conclusions: our findings indicate the high incidence of PDGFR α and β immunoexpression in the trephine biopsies of MM patients and most importantly underline the correlation between the high PDGFRβ expression with the advanced MM.
