Summary:
The present Doctoral thesis, which is developed in 5 chapters, focuses for the first time on the phytochemical study of 11 taxa (10 species, 2 subspecies) of the genus Helichrysum of Greek origin and in particular the species H. amorginum.
Chapter 1 introduces the family Asteraceae and the genus Helichrysum, referring to botanical, phytochemical, and ethnopharmacological information of the species of the genus. This information is then focused on the 11 Greek native taxa (six endemic or subendemic) that are found as wild-growing in Greece.
In Chapter 2, the qualitative analysis of methanolic extracts of 11 Greek taxa of Helichrysum Mill. sourced from the wild is described using the UPLC-HRMS technique. The chemical categories of the metabolites designated by the specific methodology are described with comments for each wild taxon.
In Chapter 3, a case study is carried out regarding one of the 10 species of Greek origin, namely H. amorginum. Qualitative analysis of essential oils of nine ex-situ cultivated H. amorginum genotypes from the Institute of Plant Breeding and Genetic Resources, Hellenic Agricultural Organization Demeter (ELGO Dimitra), Thermi - Thessaloniki was performed, based on gas chromatography coupled with a mass spectrometer (GC-MS). Hierarchical Cluster Analysis (HCA) and Principal Component Analysis (PCA) were employed to identify differences/similarities between the essential oils (between groups) among the 10 samples of the nine genotypes as well as a principal component analysis in percentages (%) of the nine main components of the 10 samples of the nine genotypes.
In Chapter 4, a metabolomic analysis is carried out using the UPLC-HRMS technique regarding 19 genotypes of H. amorginum ex-situ cultivated at the Institute of Plant Breeding and Genetic Resources, Hellenic Agricultural Organization Demeter (ELGO Dimitra), Thermi - Thessaloniki. Methanolic extracts were prepared and analyzed with the UPLC-HRMS technique and a metabolomic model was applied to distinguish the qualitative composition of extracts based on the identification of specific metabolites - biomarkers which are responsible for the seasonal variation of their metabolic profile.
Finally, in Chapter 5, a qualitative analysis of extracts was carried out using the GC-MS, UPLC-HRMS technique, as well as the isolation of secondary metabolites from organically cultivated H. amorginum by the company Korres Natural Products SA in its original native range in Amorgos Island. Also, the DPPH free radical scavenging activity as well as tyrosinase enzyme inhibition of H. amorginum extracts were studied.
