The increasing prevalence of pharmaceuticals, personal care products, and other anthropogenic substances in wastewater highlights the need for advanced analytical methods to monitor these biomarkers. Wastewater-Based Epidemiology (WBE) treats wastewater as a composite urine sample, rich with various chemicals that indicate human exposure to different substances. By monitoring these chemicals, WBE can offer comprehensive insights into the health and lifestyle habits of a community. However, the chemical analysis of wastewater is a significant challenge for analytical chemistry due to the complex nature of the matrix and the typically trace levels of biomarkers present.
In most WBE studies, liquid chromatography-tandem mass spectrometry (LC-MS/MS) has emerged as the predominant technique for targeted biomarker analysis due to its sensitivity, which is essential for detecting compounds at low concentrations. However, there are few comprehensive multi-component methods (>100 analytes) available in the literature for determining a wide range of compounds in wastewater.
This thesis presents the development of a novel multi-component method for surveilling biomarkers in wastewater utilizing liquid chromatography-tandem mass spectrometry (LC-MS/MS). The selected biomarkers include various pharmaceutical classes and their metabolites, reflecting the pharmaceutical market in Greece. Additionally, key drugs of abuse and transformation products of highly unstable compounds in wastewater are included.
A generic mixed-mode solid-phase extraction protocol was employed for the pretreatment of influent wastewater samples. The resulting extracts were analyzed using a UPLC-MS/MS system (Sciex Qtrap 5500) with a scheduled multiple reaction monitoring (sMRM) scan mode method, achieving a short chromatographic run time of 14 minutes.
